How to read a peptide Certificate of Analysis: a field-by-field reference

Research-use-only. This is an analytical reference to the fields on a synthetic-peptide certificate of analysis and what each one means. It makes no claims about biological activity, dosing, safety, or human use. The cleanliness screens described (endotoxin, microbial) characterize the material; they do not qualify it for any use, and nothing here should be read as implying otherwise.

What this is. A certificate of analysis (COA) answers two separate questions — is this the right molecule (identity) and how much of it is here, and how pure (content and purity) — and a rigorous one documents both, discloses its methods, and can be verified against a specific lot. This table defines each field, the method behind it, what a rigorous entry looks like, and the red flag that marks a weak one.

How to use it

Read past the headline percentage to the structure underneath. The two questions a COA must answer are independent — identity (mass spectrometry) and purity/content (HPLC + mass balance) — and no single field answers both. A strong certificate discloses its methods, reports net peptide content and not just an area-%, accounts for the non-peptide mass (counter-ion, water), screens for contamination, and is tied to a verifiable lot. The absences are as telling as the entries.
